Modacare 300

The active ingredient in the wakefulness-promoting drug Modacare 300 mg is modafinil. It is mainly used to treat excessive drowsiness caused by disorders such as shift work sleep disorder, narcolepsy and obstructive sleep apnea. Modafinil selectively promotes alertness and wakefulness, thus enabling individuals to remain awake and functional during the day, compared to traditional stimulants. 

MECHANISM OF ACTION 

Unlike amphetamines, which cause broad, non-selective CNS stimulation, modafinil appears to selectively activate specific wakefulness-related brain regions particularly the hypothalamus and parts of the brainstem rather than stimulating the entire CNS. Modafinil weakly binds to and blocks the dopamine transporter (DAT), the protein responsible for clearing dopamine out of the synaptic cleft. With DAT inhibited, dopamine builds up in the synapse, leading to stronger stimulation of postsynaptic dopamine receptors. Modafinil activates orexin (hypocretin)-producing neurons in the lateral hypothalamus, a key system that regulates arousal and wakefulness.

DOSAGE
